Biotra – LC Pro is a separation technique in which the mobile phase is an aqueous solution or an organic solvent that is miscible with water, and the compounds of interest are dissolved in water or an organic solvent such as methanol. The major advantage of the technique is the flexibility it allows in choosing the stationary and mobile phases that best separate the particular sample components. More than half of the types of columns used in Biotra – LC Pro   is reverse phase; around a quarter are normal phase; about 14% are ion-exchange columns. Obtaining high resolution at high flow rates is the key requirement of any Biotra – LC Pro   system. Separation of the different components of a mixture is achieved by allowing the sample to migrate through the column at selected experimental conditions. Discrete bands are formed because the different components of the mixture migrate through the column at different rates.

The apparatus of a typical liquid chromatography system includes an fluent system, a pump, an automatic sampler, the analytical column, a detector and a recorder. The most essential component of this apparatus is the analytical column that is used to separate the components of interest in biological samples. This is a sensitive detector for organic molecules that absorb light within the UV or visible wavelength spectrum, which is widely applicable to many compounds. Sample preparation also involves the typical methods of solvent extraction, ultra filtration or solid-phase extraction. With total automation, time spent on sample extraction and clean-up procedures are minimized, while the sensitivity and simplicity of the technique are maintained. This has proven extremely useful in the analysis of catecholamines and in determination of amino acid metabolism.

ELSD

An ELSD (EVAPORATIVE LIGHT-SCATTERING DEDECTOR) atomizes the column cluate, shines light on the resulting particulate components and detects the resulting scattered light. Theoretically, an ELSD can detect any nonvolatile component. An ELSD has sensitivity roughly 10 times higher than an RI detector, but has a slightly low sensitivity to low molecular components due to theor small size. An ELSD is used mainly to detect non-UV – absorbing components. Attention should be given to the fact that nonvolatile salts cannot be used as the fluent.

Fluorescence detectors are probably the most sensitive among the existing modern HPLC Detectors its possible to detect even a presence of a single Analytes molecule in the flow cell. Typically fluorescence sensitivity is 10- 1000 higher than that of the UV detector for strong UV absorbing materials. Fluorescence detectors are very specific and selective among the others optical detector. This is normally used as an advantage in the measurement of specific fluorescent species in samples

The detection of any component significantly depends on the chosen wavelength and if one component could be detected at 280 ex and 340Em, another could be missed most of the modern detectors allow fast switch of the excitation and emission wavelength. DAD / PDA

Photo diode arrays (semiconductor device) are used in the detection unit. A DAD detectors the absorption in UV to VIS region. While a UV-VIS has only one sample –side Light receiving section a DAD has multiple (1024 for 102455/2455U) Photodiode arrays to obtain information over wide range of wavelengths at one time which is a merit of the DAD

RI DETECTOR

An RI detector detects components based on the refraction of light in solution. The flow of RI detectors is divided into the sample – side and reference –side cells. Prior to analysis, elute is introduced into either cell. Until the flow of the elute becomes equilibrated. The reference-side cell is filled with elute and the column elute is introduced into the sample-side cell through the changed flow channel. When components are eluted from the column the chemical composition changes in the sample-side solution. Which changes its photorefractive level as a result, the amount of high received by the light-receiving section changes, showing a peak which can be detected. Any component in the elute can be detected. Thus, the RI detector is often called a “universal detector”.

PDD

PDD has two different modes one is PDECD and another one PDHID. The PDECD is a selective detector for monitoring high electron affinity compounds such as Freon, chlorinated Pesticides and other halogen compounds for this type of compound the minimum detectable quantity (MDQ) is at the femtogram (10 – 15)   or pictogram (10-12) level. The PDD is similar in sensitivity and response characteristics to a conventional radioactive ECD and can be operated at temperature up to 400 c.

For operation in this mode He and CH4 are introduced just upstream from the column exit. The other mode PDHID is an universal non destructive high sensitivity detector. The response to both inorganic and organic compounds is linear over a wide range response to fixed gases is a positive (increasing in standing current) with an MDQ in the low PPb range.
